Apoptosis Induction by Curcumin in Human Myelogenous Leukemia Cell Lines

Abstract
Background: Curcumin, a natural, biologically active compound extracted from rhizomes of Curcuma species, has been shown to possess potent anti-inflammatory, anti-tumor and anti-oxidative properties. We investigated effects and possible mechanism of this agent on 2 human myelogenous leukemic cell lines.

Methods: K562 and KG-1 cells were selected. MTT assay was used for cytotoxicity and flow cytometry for cell cycle analysis. Protein expressions were analyzed by Western blotting and caspase activity was checked.

Results: Two cell lines showed dose-dependent susceptibility to curcumin. Cell cycle analysis showed increased sub-G1 phase in KG-1 cell. In K562 cell, PCNA (proliferating cell nuclear antigen) and cyclin D1 and B1 were down regulated by curcumin. Akt was also down-regulated and caspase-3 was activated to induce cleaved PARP (polyadenosine ribose polymerase) and apoptosis. However, phospho-Erk was not affected. Co-treatment of cyclosporin A (CsA) with curcumin resulted in attenuation of apoptosis in K562 cell, implying that curcumin-induced apoptosis depends on the cytochrome c release from the mitochondria.

Conclusion: Curcumin induced cell cycle arrest and apoptosis in human myelogenous leukemic cell lines and the apoptosis seemed to be dependent on the release of cytochrome c from the mitochondria.
